ELOVL6 promoter binding sites directly targeted by sterol regulatory element binding protein 1 in fatty acid synthesis of goat mammary epithelial cells

The elongation of long-chain fatty acid family member 6 (ELOVL6) gene plays an important role in the synthesis saturated and monounsaturated acids. Although some studies have revealed that ELOVL6 is target sterol regulatory element binding protein 1 (SREBP1; name SREBF1) rodents, mechanism underlying regulation during lactation dairy goats remains unknown. present study aimed to investigate transcriptional goat mammary epithelial cells (GMEC). We used PCR clone sequenced a 2,370 bp fragment 5? flanking region from genomic DNA. Deletion analysis core promoter located ?105 ?40 upstream start site. Mutant elements (SRE) 3 significantly reduced activities GMEC. Both SRE1 SRE3 sites were required for basal activity ELOVL6. Luciferase reporter assays showed SREBF1 knockdown decreased Furthermore, simultaneously mutated completely abolished stimulatory effect repressive linoleic on activities. chromatin immunoprecipitation confirmed SREBP1 directly bound SRE promoter. In conclusion, these results indicate regulates transcription via gland.
